May 4, 2010 ? Researchers at the Swedish medical university Karolinska Institutet have discovered a new control mechanism in our immune system. The discovery is of potential significance to the treatment of serious diseases such as MS (multiple sclerosis), rheumatoid arthritis, and SLE (Systemic lupus erythematosus).
"Now that we've started to understand the regulatory mechanisms involved in these autoimmune diseases, we are hopeful that new treatments can be found," says Mikael Karlsson, associate professor at the Department of Medicine at Karolinska Institutet in Solna, and one of the team behind the study now published in the highly reputed periodical, The Journal of Experimental Medicine.
An important component of our immune defence is a type of cell called a B cell. Normally, the job of these cells is to produce antibodies, which in turn bind to and neutralise invasive microorganisms, such as bacteria and viruses. In people with an autoimmune disease, explains Dr Karlsson, these B cells actually have an injurious effect and instead of serving the body, are activated against its own tissues, which they start to break down.
Patients with SLE and other autoimmune diseases have lower levels of so-called NKT cells. Previously, it was not known what part these cells play in the origin and development of the disease; now, however, the research group at KI has shown that this deficiency is a contributory pathogenic factor.
"We've demonstrated that NKT cells can regulate how B cells become activated against healthy tissue, and that a lack of NKT cells results in greater misguided B cell activation," says Dr Karlsson. "So now we can mechanically link the NKT cell defect in patients to the disease."
The study also shows that the NKT cells directly impede faulty B cell activation, and that they do so early in the misdirected process. The team managed to inhibit the activity of pathogenic B cells by adding NKT cells -- a result that may one day lead to new types of treatment.
"This means that new treatments specifically targeting the protective NKT cells can help this patient group," concludes Dr Karlsson.

BALTIMORE (AP) ? The Navy on Wednesday identified two sailors who perished during routine diving operations at a test pond at an Army site in Maryland.
Officials said 28-year-old Navy Diver 1st Class James Reyher of Caldwell, Ohio, and Navy Diver 2nd Class Ryan Harris of Gladstone, Mo., died Tuesday. They were assigned to Mobile Diving and Salvage Unit 2 at Joint Expeditionary Base Little Creek-Fort Story in Virginia Beach, Va.
The two had been diving at the Aberdeen Proving Ground in northeastern Maryland. One diver was pronounced dead at the scene, the other was transported to a local hospital, where he was pronounced dead later.
Tuesday's incident at Aberdeen's Underwater Test Facility occurred less than a month after George Lazzaro Jr., a 41-year-old engineering technician, died while performing maintenance at the same site.
Kelly Luster, a spokesman for the Aberdeen Proving Ground, said Tuesday's deaths are not believed to be connected to the earlier incident. But officials said Wednesday that the test facility has been closed for all diving operations until investigations into the recent deaths are done.
The test facility, dubbed the "Super Pond," is used to conduct shock testing of vessels, submarine systems and munitions. With a bottom measuring 300 feet in diameter and a maximum depth of 150 feet, the facility also has been used in testing torpedoes, missiles, warheads, amphibious and remotely controlled vehicles, underwater gun firing and acoustics.
The sailors belonged to a unit whose salvage operations have included TWA Flight 800, Swiss Air Flight 111, the space shuttles Challenger and Columbia, and the Civil War ironclad USS Monitor.
The unit also provided damage assessments and repairs on the USS Cole and participated in humanitarian missions following Hurricane Katrina and the 2010 earthquake that struck Haiti.
Deputy Commander Capt. John Coffey said Wednesday, "Petty Officer Harris and Petty Officer Reyher were exceptional Sailors."
CARACAS, Venezuela (AP) ? Venezuela's vice president said Thursday that Hugo Chavez is still fighting for his life, yet a recent poll says nearly three in five Venezuelans believe their president will return to power.
Nicolas Maduro, Chavez's self-appointed successor, said on television that his boss "is battling there for his health, for his life, and we're accompanying him."
The vice president had characterized Chavez's condition similarly on Dec. 20, saying the president "is fighting a great battle ... for his life, for his health."
Chavez hasn't spoken or been seen since before his fourth operation in Cuba on Dec. 11 for an unspecified cancer in the pelvic area.
The government says he has been breathing with the help of a tracheal tube after surviving a serious respiratory infection. It says Chavez returned on Feb. 18 and is at a military hospital in Caracas for continued treatment for "respiratory insufficiency."
Despite speculation by doctors not involved in Chavez's treatment that it is most likely palliative, designed only to make him more comfortable in his remaining days, many Venezuelans apparently believe ? or want to believe ? he is on the mend.
"The president's prolonged absence and his critical situation have not been converted into massive pessimism about his return," respected pollster Luis Vicente Leon tweeted Thursday.
He said nearly 58 percent of Venezuelans believe Chavez will recover while about 30 percent believe he will not return to power and 12.5 percent say they don't know what will happen. One percent, meanwhile, believe Chavez was never sick.
Leon, chief of the Datanalisis polling firm, told The Associated Press that the Feb. 11 poll of 1,198 people had an error margin of three percentage points.
He said he thought the poll reflected people's desire not to believe the worst about someone who is dear to them, just as people resist accepting that a close relative might be dying.
Leon also said he thought reports of government officials holding hours-long meetings with Chavez had contributed to the belief of many Venezuelans that Chavez will return.
"The government has sent permanent messages that President Chavez will return, that he meets with the vice president for five hours," Leon noted.
He said people don't necessarily believe that, however, as the poll found 44 percent think the government has not been transparent in discussing the Chavez's health.
In his televised remarks, Maduro called for Venezuelans to keep praying for Chavez and remain loyal to the committed socialist who has been their president for more than 14 years.
"Do you know why Comandante Chavez neglected his health and has been battling (cancer) for nearly two years?" he said. "Because he completely surrendered body and soul and forgot all his obligations to himself in order to give himself to the homeland."
Chavez, 58, himself has previously acknowledged that he was neglecting his health in recent years, often staying up late and consuming cup after cup of coffee to remain alert.
The president has undergone surgeries, chemotherapy and radiation treatments since June 2011, when he first announced his cancer diagnosis. He hasn't specified the type of cancer or the exact location in his pelvic region where his tumors have been removed.
On Feb. 15, the government released four photographs of Chavez lying in a bed in Cuba with his two daughters by his side. They were the only images of him published since early December.
Re-elected in October, Chavez was scheduled to have been sworn in on Jan. 10. But the Supreme Court said the swearing-in could be delayed.
NEW YORK (AP) ? Idina Menzel, who won a Tony Award as the green witch in "Wicked," is planning a return to Broadway.
Producers of a new musical by the creators of the Pulitzer Prize-winning "Next to Normal" announced Thursday that Menzel will star in their "I/Then," a Broadway-bound musical that first takes a stop this fall in Washington, D.C.
Tom Kitt will supply the music, Brian Yorkey will write the story and lyrics, and Michael Greif will direct. All three did the same jobs for "Next to Normal."
The new show will make its world premiere this November at the National Theatre in Washington, D.C., and then go on to open on Broadway in March 2014.
Producers said the musical will center on the character of Elizabeth, who on the verge of turning 40, moves to New York. "Set against the ever-shifting landscape of modern day Manhattan, 'If/Then' is a romantic and original new musical about how choice and chance collide and how we learn to love the fallout," producers said in a statement.
Menzel made her Broadway debut in "Rent" and won a Tony as Elphaba in "Wicked." Her films include "Enchanted," ''Rent" and "Ask the Dust." She also has launched a solo recording career and had a recurring role on TV in "Glee."

First fossil evidence shows small crocs fed on baby dinosaurs
RAPID CITY, S.D. (Feb. 28, 2013) A South Dakota School of Mines & Technology assistant professor and his team have discovered a new species of herbivorous dinosaur and published the first fossil evidence of prehistoric crocodyliforms feeding on small dinosaurs.
Research by Clint Boyd, Ph.D., provides the first definitive evidence that plant-eating baby ornithopod dinosaurs were a food of choice for the crocodyliform, a now extinct relative of the crocodile family. While conducting their research, the team also discovered that this dinosaur prey was a previously unrecognized small ornithopod dinosaur species, which has yet to be named.
The evidence found in what is now known as the Grand Staircase Escalante-National Monument in southern Utah dates back to the late Cretaceous period, toward the end of the age of dinosaurs, and was published Feb. 27 in the academic journal PLOS ONE (Public Library of Science ONE). The complete research findings of Boyd and Stephanie K. Drumheller, of the University of Iowa and the University of Tennessee, and Terry A. Gates, of North Carolina State University and the Natural History Museum of Utah, can be accessed at http://dx.plos.org/10.1371/journal.pone.0057605.
A large number of mostly tiny bits of dinosaur bones were recovered in groups at four locations within the Utah park which paleontologists and geologists know as the Upper Cretaceous (Campanian) Kaiparowits Formation leading paleontologists to believe that crocodyliforms had fed on baby dinosaurs 1-2 meters in total length.
Evidence shows bite marks on bone joints, as well as breakthrough proof of a crocodyliform tooth still embedded in a dinosaur femur.
The findings are significant because historically dinosaurs have been depicted as the dominant species. "The traditional ideas you see in popular literature are that when little baby dinosaurs are either coming out of a nesting grounds or out somewhere on their own, they are normally having to worry about the theropod dinosaurs, the things like raptors or, on bigger scales, the T. rex. So this kind of adds a new dimension," Boyd said. "You had your dominant riverine carnivores, the crocodyliforms, attacking these herbivores as well, so they kind of had it coming from all sides."
Based on teeth marks left on bones and the large amounts of fragments left behind, it is believed the crocodyliforms were also diminutive in size, perhaps no more than 2 meters long. A larger species of crocodyliform would have been more likely to gulp down its prey without leaving behind traces of "busted up" bone fragments.
Until now, paleontologists had direct evidence only of "very large crocodyliforms" interacting with "very large dinosaurs."
"It's not often that you get events from the fossil record that are action-related," Boyd explained. "While you generally assume there was probably a lot more interaction going on, we didn't have any of that preserved in the fossil record yet. This is the first time that we have definitive evidence that you had this kind of partitioning, of your smaller crocodyliforms attacking the smaller herbivorous dinosaurs," he said, adding that this is only the second published instance of a crocodyliform tooth embedded in any prey animal in the fossil record.
"A lot of times you find material in close association or you can find some feeding marks or traces on the outside of the bone and you can hypothesize that maybe it was a certain animal doing this, but this was only the second time we have really good definitive evidence of a crocodyliform feeding on a prey animal and in this case an ornithischian dinosaur," Boyd said.
The high concentrations of tiny dinosaur bones led researchers to conclude a type of selection occurred, that crocodyliforms were preferentially feeding on these miniature dinosaurs. "Maybe it was closer to a nesting ground where baby dinosaurs would have been more abundant, and so the smaller crocodyliforms were hanging out there getting a lunch," Boyd added.
"When we started looking at all the other bones, we starting finding marks that are known to be diagnostic for crocodyliform feeding traces, so all that evidence coming together suddenly started to make sense as to why we were not finding good complete specimens of these little ornithischian dinosaurs," Boyd explained. "Most of the bites marks are concentrated around the joints, which is where the crocodyliform would tend to bite, and then, when they do their pulling or the death roll that they tend to do, the ends of the bones tend to snap off more often than not in those actions. That's why we were finding these fragmentary bones."

In the process of their research, the team discovered through diagnostic cranial material that these baby prey are a new, as yet-to-be-named dinosaur species. Details on this new species will soon be published in another paper.
